Synaptic adhesions are critical components for synaptogenesis. Han et al. identify physical interactions of neurexins with leukocyte common-antigen related receptor tyrosine phosphatases (LAR-RPTPs) that are regulated by availability of heparan sulfates and alternative splicing of LAR-RPTPs. Neurexins and LAR-RPTPs cooperate in presynaptic assembly, whereas they competitively function in postsynaptic differentiation. Drosophila orthologs of neurexins and LAR-RPTPs control structure and function at neuromuscular junctions. This study sheds light on how trans-synaptic signals bidirectionally orchestrate synapse organization.
